Uranium and/or plutonium should be in the game, with both an unrefined and refined version. It could be found submerged in lava in the oil biome, or in small amounts on asteroids. Beyond that, some rare/special critter types could produce it, with the additional problem of the critter spreading radiation around.

The refined version should be fuel for a massive nuclear power building with a huge heat output and the ability to be partly submerged in water for cooling. Refined radioactive material might also be necessary components for various other late-game ideas, such as teleporters, or even an end-game spaceship structure, etc.

Radiation and radiation should be easy to add. You can use the existing disease/bacteria system, and make it where the radiation can't be "killed" by anything other than a special filtering building. The primary source of radiation would be radioactive material itself in addition to radioactive waste put out by the nuclear power plant.

Bunker walls should be the only tile capable of preventing radiation from creeping through.

In my opinion the nuclear reactor should just produce a lot of heat and not generate any power, perhaps it could even consume a little bit of power. The heat it produces would be used to power one or more steam turbines.

Yea everyone expects Nuclear reactors in the future.  Likely as heat producers that you then need to connect to a Steam Generator.

Radiation is not well modeled as a germ type.  Radiation dosage is cumulative without any safe level, radiation travels through empty space.  So I would recommend something more like the light system in which radiation is a emanating from a source and has a 'rad' value in space that declines with distance and from mass shielding it.
