It would actually be fairly easy to test, as puzzle two can provide you the information to be certain what the placement must be before you "confirm" your solution and complete part two. Given the visual feedback, my guess would be that completing stage two is necessary to unlock step three. But I'd be interested to know if anyone has confirmation one way or another.
