DragonDePlatino
I'd say it's just an artistic choice. One of the aspects of Don't Starve's style is that characters don't have pupils. This means that most of the characters either have huge ghostly eyes or beady cartoon eyes. I actually do that in my character designs too. (ooold art)
YouCleverBoy
Because they have no souls. Simple. I mean, you have to have no conscience to burn down a whole forest with no remorse, summon your dead sister back from the grave, slay creatures and steal their life essence, or be totally cool with living as a spidery abomination of nature. And the animals, well they live in a nightmarish wilderness. They have no souls either.

Mobbstar
Because of artistic choice, as everyone knows already. But why exactly? Blank eyes take some humanity away, and create an eerie feeling. Willow and Wendy fit with that description, the other characters don't. Also, pupils point towards a specific direction, whereas blank (or button-) eyes only indicate whether the character is looking at all. The latter is perfect for the wildlife of DS, as it has four directional animations, but the player can be anywhere within those four sectors (not the mathematically correct word, but whatevs). Pupils would make the rabbits and stuff look really stupid and seemingly looking elsewhere, a great problem for new players, and an annoyance to everyone. Klinx
Honestly, I believe it's just a choice of art. I love it. I draw all kinds of strange creatures in my own art and they always lack in pupils. It just gives this look of emptiness and mystery.
